The usual instructions for opening all the depth 5 FRC dungeon glyphs we use to farm blood gems are: First, finish all the story dungeons (skipping Hintertomb), which gives you all the depth 5 root chalices. Then, create any one depth 5 FRC root dungeon of your own: an FRC Isz requires the fewest materials. To open a dungeon, you need to own a root chalice for that dungeon type.Sinister root chalices “count” as root chalices. Many in the community know one or both of these, but for those who don’t, here you go: And this is totally true! But there are a couple shortcuts that are sometimes helpful. After that, you can open any depth 5 FRC glyph. But sinister root chalices will do the trick. You yourself must create at least one dungeon with F, R and C offerings. If you’re rushing to get that uncanny Burial Blade in a Lower Loran root and wish you could skip the Darkbeast… you can! (The sinister trick doesn’t help with Pthumeru Ihyll, since the root chalice drops from the layer 2 boss, before you can get the sinister version.) This can be a time-saver because the Lower Loran Root and Isz Root Chalices drop from the layer 3 bosses, Loran Darkbeast and Ebrietas, but the Sinister versions can be bought from the messengers earlier on layer 3, without having to face those bosses. So you can open an Isz Root dungeon if you have the Sinister Isz Root chalice, even if you don’t have the regular Isz Root Chalice. To join a glyph with one or more of the special offerings Fetid, Rotted and Cursed, the game checks to make sure you have made at least one dungeon of your own with each offering on the glyph.BUT it doesn’t have to be all the same dungeon, and it doesn’t have to be depth 5.

Hunters can access these multi-leveled dungeons by performing a Chalice Ritual. Root chalice dungeons are procedurally-generated, whereas other chalice dungeons are fixed in structure. Although the dungeons' layout changes with each ritual performed, each layout is saved and can even be uploaded and shared with friends or the world.
